almost all sample preparation techniques kill specimens. you are going to see nothing fluorescent under a light microscope, stains kill things, prep for electron microscopy kills things. What r u left with?

The microscope technique that would allow you to see specific parts of living specimen is the stain process. You will be able to visually see the different structures of a specimen. This is because the specimens are colorless, that you wouldn't be able to see the structure without a stain. Hope that helps! n_n

But hector is right: stains which don't kill the specimen are few and far between. It says "living" in the info. Passing refracted light from the specimen through a pinhole is called "dark field microscopy"...

Staining is the easiest and best way for observing a specific part of living organisms.... With this you can also use fluorescent dye and phase contrast microscopy but in phase contrast observation of a living organism is not possible...... and in dark field microscope you can't see a specific part of an organism....
