What is the difference when you write f[x]=x+3 instead of f[x]:=x+3 in Mathematica?

I dont think f[x]:=x+3 will work

f[x]=x+3 give 3+x

f[x]:=x+3 gives nothing

As far as I know, in mathematic, the following function will give erro f[x]:=x+3 Becase It would assume them as ratio and you cannot have an = sign in denominator
