It was geographically suited for bombing and evaluating the results of bombing. It would have few casualties because it was nearly destroyed by previous bombings. It was a religiously and culturally significant city for the Japanese monarchy. It was a city with many family connections to Emperor Hirohito and military leaders.

Hiroshima was a city that had a well-sized population, but the people that lived there weren't central to Japan - ie no familial ties, cultural significance. Really, the Americans bombed Hiroshima because it would set a precedent or example for the rest of the Japan and especially Hirohito - telling them that 'this is what the Americans can do, we should back off'
