The most significant reason for the assassination was because Booth believed that it would allow the Confederate states to gain momentum and revive their cause

Booth was a Confederate sympathizer who saw Lincoln as a tyrant. He saw the assassination as a blow for liberty, and therefore carried it out in public for all to see, and so that he himself would go down in history. The assassination of Lincoln was part of a wider plot; the Vice-President and Secretary of State were also supposed to be assassinated at the same time.
