what is a limitation of using electron microscopes to view specimens?

OpenStudy (anonymous):

the specimen has to be dead and wont be in colour only black and white and they will be only small ones we cant view large specimens

Actually you can view large specimens, something as big as a drosophilia if you like as long as it's an SEM. Other limitations: The Scanning Electron Microscope (SEM) can only view 3D surface texture of objects. It bounces electrons off of your sample. So you can't view internal structures. Plus it is limited to about \[10^{-9}m\]. Transmission electron microscopes allow you to see through structures as it emits electrons powerful enough to pass through the specimen, with a resolution limit of \[10^{-11}m\]
