friends....IF THE SPECIMENS WE WANT TO OBSERVE ARE NOT TRANSPARENT THAN HOW TO SEE THEM THROUGH MICROSCOPE?

hi @rusbhara ..let me know about the type of specimen you are looking at..because microscopic techniques depend on what actually you want to observe

@biocrystal...i m studying microbiology now first year...so our sir asked us question that if a specimen is not transparent then ow to see it through microscope? now the specimen might be the colonies of different kinds of bacteria..so help me here

a phase contrast microscope helps to look at such things..try reading more about phase contrast microscopy

there will be some media ' s like saffranin, crystal blue that give colour to the specified part of the bacteria which helps us to learn them

but being not transparent itself is the main thing one needs for better study especially in microbiology..most of the times bacteria being transparent causes a great problem to observe under microscope..that is why we use stains to create a phase difference and make it easily observable with human eye under microscope..

that's right @deepika.comet. but the question here is not about transparent things.. so transluscent things are already good enough to be seen under a phase contrast microscopy without any staining
