Hello, just want to ask if there any diferences between Gram - and Gram + in term of rate of decolorization? (Gram staining exp) i really appreciate answer from all of you

@syay43 It is not about the rate really. The reason the stain works is because of the differences in the thickness of the cell wall. Once the gram+ bacterium are stained, the stain cannot be easily removed with the chemical that does the destaining. Gram- bacterium are easily destained.

@syay43 This diagnostic can be difficult to do well. Technically the counter stain for gram- is not necessary. If the stain can be removed it is gram-. Another issue is that excessive destaining can damage the cell walls enough to ruin the results. Finally, some bacteria, like acid-fast bacteria, have a waxy coating that can confuse the reading of a gram stain.

oh..it ok actually our lab assistance already prepared specific bacteria.. hehe thnks a lot @mrdoldum you really a big help to me. about acid fast bacteria.. i use Mycobacterium phlei.

@syay43 Yeah, when I was a teacher's assistant for Microbiology Mycobaacterium really confused students on the gram stains.

@syay43 I prefer to not to give out such information due to the amount of geographical knowledge that would provide. However, I was a TA for 7 lab sections of Microbiology when I did my BS in Biology. I am currently in the last semester of my PhD in a molecular biology field, and teach intro bio labs at this moment.
