To Identify perfect square trinomials. The first term is a positive perfect square. Last term is a positive perfect square, and the middle term is twice the product of the square root of the first term and the square root of the third term.

A binomial is a Difference of Squares if both terms are perfect squares. A^2 - B^2 = (A - B)(A + B) So for either of these two yes, we have a difference of squares. x^2 - 25 16^4 - 100 but this one 50^2 - 72 neither 50^2 or 72 are perfect squares so you wouldn't have a different of squares.
