During DNA replication the helicase actually separates the strands of DNA from each other by breaking the hydrogen bonds between base paris. @shreehari499 I would be cautious to say that the helicase uncoils the DNA as there are many levels of DNA coiling in the nucleus, and in this case we are specifically talking about the separation of the two strands. In fact enzymes such as topoisomerase, which change the level of DNA supercoiling actually make cuts in the DNA wherease the helicase does not (during replication).
