You start with a bunch of homologous DNA molecules. You treat them with a restriction enzyme which "digests" the DNA molecules by recognizing short sequences in the DNA and cleaving the backbones at those sites. You then run the fragments on a gel to separate them by size and transfer them to a membrane by southern blotting. Then you treat the membrane with a probe sensitive for DNA fragments of different lengths and see what binds. A RFLP occurs when the length of a detected fragment varies among individuals. This was a long time ago for me.
