.What is a limitation of using electron microscopes to view specimens? Resolution is weak and cannot make out details of very small specimens. Electron beams are dangerous and can only be used sparingly. You cannot view live specimens because the necessary preparation kills cells. You cannot view transparent specimens because of the microscope’s low resolution.

Preparation for electron microscopy kills the specimens so you can't see actual living processes taking place. For those interested, quantitative confocal microscopy provides the best high resolution visualisation of living processes out there. It is puzzling that intro biology courses don't touch on it more often as so many biologists spend so much of their careers using it...
