It doesn't really matter. Descartes' rule of signs is optional, it can tell you whether to look for positive or negative roots first, but you need rational root theorem to know exactly where to look. DRS might narrow it down sooner than RRT, but in practice, it doesn't really matter.

If you really had to choose between the two, I'd choose DRS, but it really doesn't matter. You ought to do both before you go looking for roots, so as long as both get done, it doesn't make a difference which gets done first. RRT has to be done. DRS is more like icing on the cake.
