when you perform a southern blot, should you know about the sequence of the desired gene prior to inserting the probe?

heya...according to what i know...southern blot is carried out to find out or detect if there is the presence of a specific nucleotide sequence in d isolated genome. so yes you should know d sequence. plus the probe is complementary to the sequence so to make that probe you need to know d sequence. i also found this on mcgraw hill... http://highered.mcgraw-hill.com/olcweb/cgi/pluginpop.cgi?it=swf::535::535::/sites/dl/free/0072437316/120078/bio_g.swf::Southern+Blot
