This method is used to determine the amino acid sequence of a template DNA. A typical automated sequencing reaction using fluorescence consists of a DNA template, four dNTPs (dATP, dCTP, dGTP, dTTP), a primer, buffer components, DNA polymerase, and four different fluorescently-labeled ddNTPs (ddATP, ddCTP, ddGTP, ddTTP).
